Purpose: The purpose of the lab is to determine the effects of low, medium, and high doses of an energy drink to the growth of the Yeast Strain S. cerevisiae. The lab will then help us identify the effects that energy drinks have on the body.
Hypothesis: Based on the fact that we are mixing energy drinks with yeast and past studies I would expect that the yeast will decrease the growth based on all the harmful ingredients that is being mixed into the yeast.
Brief Protocol:
Preparing the Energy Drink:
- We will begin by diluting the amount of energy drink that we are going to mix with the yeast to in order to compare a low, medium, and high dose of the energy drink:
.5 ml energy drink with 4.5 ml water
1 ml energy drink with 5 ml water
2 ml energy drink in 3 ml of water
Procedure:
- Place the yeast into 5ml of water to use as our control experiment to observe the average growth.
- Place the yeast into the different solutions of the energy drink.
- Repeat step 1 and 2: 3 times in case there is a variable that affects the growth of the growth.
- Leave inside the different solutions for 30 minutes each.
- Take a of 3ml of each sample and place into a cuvette.
- Record the absorbance of each solution through the spectrometer.
- Take the readings of each and use the formula: Number of cells
(number of cells/ml) x ml
- Read the number of cells based on the ml and determine if there was cell growth.
Discussion of Variables:
With our reagent being energy drinks there are many variables that could affect the outcome of the experiment. One variable that could affect the experiment is the different brands of energy drinks, each brand is made with different ingredients causing different interactions with the yeast strain. The next variable that could vary the results of the experiment is the temperature of the solutions that is being mixed into the yeast strain as the higher the temperature is the more effective the cell will be at growing and dividing. The final variable that could occur is that the amount of the energy drink or water will be added with a higher amount than what is stated.
Dilutions to be used:
I have determined to use 5ml solutions based on a previous experiment that was done using 5ml solutions mixed into the yeast. In order to test how different concentrations of the solutions affect the growth of the yeast strain. I have determined to have solutions with the majority of water, one with half water and half energy drink and the final with the majority of energy drink.
Explain your method of analysis:
For the experiment we have chosen to observe the growth of the yeast due to the fact that various ingredients within the energy drink such as caffeine can inhibit the growth of cells while other ingredients have no damaging effects. With all the ingredients combined we will try to determine how beneficial or detrimental energy drinks can be to the body. Through this method we will be able to achieve our method of analysis because with varying concentrations of the energy drinks we will be able to observe the effects whether the growth of the yeast slows down or speeds up.
