Think about what we discussed before and while doing this lab. Remember that we examined our plates three times – we looked across dilutions, across treatments, and then we counted actual colonies. To properly analyze this experiment, you should consider your data from these perspectives when discussing it.
The discussion should NOT be written as a series of answers to these questions – these ideas and considerations should be woven throughout a thoughtful, thorough discussion of the whole experiment. The plates you got are a simulation of an actual experiment; treat it as such – although you did not do the setup yourself, you can still discuss the factors that go into performing such an experiment.
Dilutions: What did our serial dilutions tell us? Why are serial dilutions so powerful and where are they most effective? How did they affect our data and counting? How did they help us answer our experimental question?
Treatments: What did we learn from our treatments? How did they affect our colony growth, and therefore our data? Were they as you predicted, or were you surprised by any results? Can you answer our experimental question? Estimates are fine, killing curves are always an educated estimation.
Colony Data: How did your data turn out? Does the overall shape of your graph make sense? If it does, what does it tell you about our experimental question? What conclusions can you draw from it? Are there any outliers? How might those have come about? Can you still draw conclusions regarding our experimental question? Can you estimate the UV dose that would result in 1% survival? What does this number tell you? If the graph does not make sense, in what way? What are some possible sources of error in this experiment? What might you do differently?
Finally, be sure to connect the bacterial mutagenesis results to genetics and biology in general.
