We will perform a cell assay to measure cell proliferation (growth) and viability. to test chemotherapy drugs for inhibition of cell proliferation of cancerous cells. The former is important for making sure therapeutic drugs can be used safely on humans.
MTT (3-(4,5-Di methyl thiazol-2-yl)-2,5-diphenyltetrazolium bromide, a tetrazole) a yellow-colored compound forms a formazan dye (purple) when reduced by reductases found in living cells. Thus it can be a reliability and sensitive method for measuring cells viability. Using are using a modified and more sensitive version a water-soluble tetrazolium salt called WST-8 [2-(2-methoxy-4-nitrophenyl)-3-4-nitrophenyl)-5-(2,4-disulfophenyl)-2H-tetrazolium, monosodium salt](Dojindo laboratories, Rockville, MD). WST-8 changes to an orange color which we can quantify by measuring absorption using the plate reader at a wavelength of 490 nm.
Protocol:
- To your treated cells
- Add 10 uL of the WST-8 to each well and incubator for 90 min.
- Make sure to examine your cells for growth and general appearance and take notes.
- Read absorbance 490 nm using the plate reader
- We in your research notebook provide data and analysis on your experiment and the class combined data as well. Need to include results.
